The study was conducted to isolate and diagnose this bacterium, identify some of its virulence factors, and determine its resistance to certain types of antibiotics, as well as to understand the variability in the expression of some virulence genes. This study included a total of 150 isolates from various sites isolated from patients in hospitals, clinics, and tuberculosis centers in Salah alDin Governorate. Among these, 55 clinical isolates, accounting for 47.82%, were diagnosed as K. pneumoniae from urine, burns, the respiratory system, and wounds through biochemical tests, with confirmation of the diagnosis using the Phytec-2 technique. To evaluate the extent of variation in gene expression of the gene (acrAB), and (recA) from the housekeeping genes. The results of the current study showed that the bacteria were resistant to most of the used antibiotics, as all isolates were resistant to the antibiotic ampicillin (AMP). Antibiotics should be developed at concentrations that affect the bacteria. The
results of the gene expression of the studied genes showed a significant increase in gene expression regulation. According to the statistical analysis, the results indicated that the acrAB gene had high gene expression with an average expression of (17.260), while the reference gene had an average expression of (20.284)
